Inhibition of the nuclear export of p65 and IQCG in leukemogenesis by NUP98-IQCG

摘要:

NUP98 fuses with approximately 34 different partner genes via translocation in hematological malignancies. Transgenic or retrovirus-mediated bone marrow transplanted mouse models reveal the leukemogenesis of some NUP98-related fusion genes. We previously reported the fusion protein NUP98-IQ motif containing G (IQCG) in a myeloid/T lymphoid bi-phenoleukemia patient with t(3;11) and confirmed its leukemogenic ability. Herein, we demonstrated the association of NUP98-IQCG with CRM1, and found that NUP98-IQCG expression inhibits the CRM1-mediated nuclear export of p65 and enhances the transcriptional activity of nuclear factor-κB. Moreover, IQCG could be entrapped in the nucleus by NUP98-IQCG, and the fusion protein interacts with calmodulin via the IQ motif in a calcium-independent manner. Therefore, the inhibition of nuclear exports of p65 and IQCG might contribute to the leukemogenesis of NUP98-IQCG.

Expression of STAT6 and NFB p65 in the colon mucosa of patients with ulcerative colitis

摘要: The expression of signal transducer and activator of transcription 6 (STAT6) and nuclear factor-κB (NF-κB) in the colonic mucosa of patients with ulcerative colitis (UC) was examined. Real-time polymerase chain reaction and immunohistochemistry were used to detect the expression of STAT6 and NF-κB p65 at both mRNA and protein levels in the colonic mucosa of patients with UC and healthy volunteers. The results showed that the expression levels of STAT6 and NFκB p65 in the colonic mucosa of patients with UC were significantly higher than in normal controls at both mRNA and protein levels. These data suggest that STAT6 and NFκB p65 perhaps play an important role in the pathogenesis of UC and underscore the potential value of anti-UC strategies in the clinical management of this disease.

Effect of compound Sophorae Flavescentis Jiechangrong capsule on expression of NFB p65 and STAT6

摘要: The effects of compound Sophorae Flavescentis Jiechangrong capsule (CSFJC) on the expression of nuclear factor-κB p65 (NF-κB p65) and signal transducer and activator of transcription 6 (STAT6) in the intestinal mucosa of patients with ulcerative colitis and the possible mechanism were investigated. Eighteen patients with ulcerative colitis were randomly divided into a traditional Chinese medicine (TCM) group ( = 11) treated by CSFJC and a western medicine (WM) group ( = 7) treated by Sulfasalazine tablets. The treatment duration lasted eight weeks. Before and after the treatment, the symptoms and the physical signs were observed, and the routine stool test, the colonoscopy, and pathological examination were performed in the two groups. The expression levels of NF-κBp65 and STAT6 were detected by using immunohistochemistry. The results showed that the total effective rate of the curative effectiveness in TCM and WM groups was 100% and 71.4%, respectively, and the total effective rate of colonic mucosa lesion in TCM and WM groups was 90.9% and 71.4%, respectively, with the differences being significant (all < 0.05). The total effective rate of syndromes of damp-heat blocking according to the TCM in TCM and WM groups was 90.9% and 71.4%, respectively. After the treatment, the expression of NF-κB p65 and STAT6 in the two groups was decreased, and the decrease of NF-κB p65 and STAT6 expression in TCM group was more significant than in WM group ( < 0.05). It was concluded that CSFJC can inhibit the activation and expression of NF-κB p65 and STAT6 in the intestinal mucosa of patients with ulcerative colitis, which is a possible mechanism for CSFJC treating ulcerative colitis.

2.5 triggers airway inflammation and bronchial hyperresponsiveness in mice by activating the SIRT2--p65

摘要: Exposure to particulate matter 2.5 (PM2.5) potentially triggers airway inflammation by activating nuclear factor-κB (NF-κB). Sirtuin 2 (SIRT2) is a key modulator in inflammation. However, the function and specific mechanisms of SIRT2 in PM2.5-induced airway inflammation are largely understudied. Therefore, this work investigated the mechanisms of SIRT2 in regulating the phosphorylation and acetylation of p65 influenced by PM2.5-induced airway inflammation and bronchial hyperresponsiveness. Results revealed that PM2.5 exposure lowered the expression and activity of SIRT2 in bronchial tissues. Subsequently, SIRT2 impairment promoted the phosphorylation and acetylation of p65 and activated the NF-κB signaling pathway. The activation of p65 triggered airway inflammation, increment of mucus secretion by goblet cells, and acceleration of tracheal stenosis. Meanwhile, p65 phosphorylation and acetylation, airway inflammation, and bronchial hyperresponsiveness were deteriorated in SIRT2 knockout mice exposed to PM2.5. Triptolide (a specific p65 inhibitor) reversed p65 activation and ameliorated PM2.5-induced airway inflammation and bronchial hyperresponsiveness. Our findings provide novel insights into the molecular mechanisms underlying the toxicity of PM2.5 exposure. Triptolide inhibition of p65 phosphorylation and acetylation could be an effective therapeutic approach in averting PM2.5-induced airway inflammation and bronchial hyperresponsiveness.

Inhibition of NF-kappa B can enhance Fas-mediated apoptosis in leukemia cell line HL-60

摘要: This study explored the effects of nuclear factor-kappa B (NF-κB) inhibitor Bay 11-7082 on Fas/FasL system and Fas-mediated apoptosis in cell line HL-60 cells. The mRNA and protein levels of Fas, FasL, and X-linked inhibitor of apoptosis protein (XIAP) were detected by reverse transcription-polymerase chain reaction (RT-PCR) and flow cytometry (FCM); the level of sFasL was evaluated by enzyme-linked immunosorbent assay (ELISA); and apoptosis was determined by FCM. After treatment with Bay 11-7082, the mRNA and protein levels of FasL and XIAP in HL-60 cells were significantly lower than in the controls ( <0.05), but the mRNA and protein levels of Fas and sFasL did not change significantly ( >0.05). Apoptotic rate of HL-60 cells treated with Bay 11-7082 was significantly higher than in the controls ( <0.05). Therefore, we conclude that Bay 11-7082 can enhance Fas-mediated apoptosis in HL-60 cells by downregulating FasL and XIAP levels.

Impact analysis of compressor rotor blades of an aircraft engine

摘要: Frequent failures due to foreign particle impacts are observed in compressor blades of the interceptor fighter MIG-23 aircraft engines in the Ethiopian air force, supplied by the Dejen Aviation Industry. In this paper, we made an attempt to identify the causes of failure and hence recommend the suitable materials to withstand the foreign particle impacts. Modal and stress analysis of one of the recently failed MIG-23 gas turbine compressor blades made up of the following Aluminum based alloys: 6061-T6, 7075-T6, and 2024-T4, has been performed, apart from the impact analysis of the rotor blades hit by a granite stone. The numerical results are correlated to the practical observations. Based on the modal, stress and impact analysis and the material properties of the three considered alloys, alloy 7075-T6 has been recommended as the blade material.

摘要:

This article describes a study on a non-thermal plasma device to treat exhaust gases in an internal combustion engine. Several tests using a plasma device to treat exhaust gases are conducted on a Honda GX200-196 cm3 engine at different rotational speeds. A plasma reactor could be efficient in degrading nitrogen oxides and particulate matter. Monoxide and carbon dioxide treatment is minimal. However, achieving 1%–3% degradation may be interesting to reduce the emission of greenhouse gases.

Bismuth oxide-related photocatalysts in green nanotechnology: A critical analysis

摘要: A survey addressing the uses of bismuth oxide in photocatalysis is presented. The richness of literature on such a specific topic proves the growing importance of this compound as a valid tool in pollution abatement and environmental decontamination. Many research groups have focused their activity on how to improve the photocatalytic properties of this semiconductor and several solutions have been adopted in the synthesis method, often based on wet-chemical processes. The impressive development of nanoscience helped in understanding and identifying process variables and operative conditions aiming at optimizing the yield of this promising photocatalytic material in the utilization of solar energy.

Inhibition of NO emission by adding antioxidant mixture in

摘要: In this paper, the effect of adding an antioxidant mixture in biodiesel as fuel, in a single cylinder, direct injection compression ignition engine was experimentally investigated and the level of pollutants in the exhaust and performance characteristics of the engine were analyzed. Nine test fuels were prepared with three antioxidants, namely, Succinimide (C H NO ), , dimethyl- -phenylenediamine-dihydrochloride (C H Cl N ), and -phenyl- -phenylenediamine (C H NHC H NH ) added to neat biodiesel at 500 parts per million (ppm), 1000 ppm and 2000 ppm and the observed experimental results were compared with those of neat biodiesel and neat diesel as base fuels. The comparison showed that NO emission was reduced drastically for the test fuels with the antioxidant addition of 2000 ppm. The maximum reduction of 10% of NO emission was observed for the antioxidant mixture in neat biodiesel, with a slight increase in unburned HC, CO and smoke opacity. In addition, the obtained experimental results reveal that the addition of two antioxidants as mixture in neat biodiesel caused improved NO emission reduction for all test fuels.

一种观点挖掘新词语权重过程性能分析 Article

摘要: 论坛和博客的普及为大量信息的处理带来了挑战和机遇。基于不同主题的信息通常包含了主观的定性词语,需要经过统计分析转换为可用的定量数据。这些数据如不恰当处理则会影响观点的正确表达。每个观点相关词的主要表义各有不同。为将词的语义转换为数据并加强对观点挖掘的分析,我们提出了一种新颖的加权方案,称为词权重推测法(inferred word weighting, IWW)。IWW通过对语境下和表义中词语重要性的计算对算法进行增强。相对已有的方法,本文提出的加权方法从分析的视角上为词语提供了合适的权重。此外,通过对包含停用词的文本分类的性能研究,提供了另一种校验方法,作为对所提出的新加权方法的补充。而通常这些停用词都会在文本处理时移除。将包含停用词这一新概念应用于本文提出的加权方法和已有加权方法,可观察到2个现象:(1)文本分类性能增强;(2)包含停用词与否,所造成的文本处理结果的差异在所提出的方法中较小,而在已有方法中较大。进而,从这2种现象得出推论。基于基准数据集的实验结果表明所提出的方法在分类精度上具有优化潜力。

A simple umbelliferone based fluorescent probe for the detection of nitroreductase

摘要: A simple nitrobenzyl-umbelliferone (NCOU1) was synthesised containing a nitroreductase (NTR) trigger moiety. The presence of NTR, resulted in the fragmentation of the parent molecule and release of the highly emissive fluorophore umbelliferone an NTR-catalyzed reduction of the nitro group. In the presence of the NTR enzyme, NCOU1 gave rise to a 5-fold increase in fluorescence intensity at 455 nm and was selective for NTR over other reductive enzymes. These results indicate that NCOU1 can be used as a simple assay for the detection of NTR.

Role of membrane and compound properties in affecting the rejection of pharmaceuticals by different RO/NF

摘要: This study was conducted to assess the merits and limitations of various high-pressure membranes, tight nanofiltration (NF) membranes in particular, for the removal of trace organic compounds (TrOCs). The performance of a low-pressure reverse osmosis (LPRO) membrane (ESPA1), a tight NF membrane (NF90) and two loose NF membranes (HL and NF270) was compared for the rejection of 23 different pharmaceuticals (PhACs). Efforts were also devoted to understand the effect of adsorption on the rejection performance of each membrane. Difference in hydrogen bond formation potential (HFP) was taken into consideration. Results showed that NF90 performed similarly to ESPA1 with mean rejection higher than 95%. NF270 outperformed HL in terms of both water permeability and PhAC rejection higher than 90%. Electrostatic effects were more significant in PhAC rejection by loose NF membranes than tight NF and LPRO membranes. The adverse effect of adsorption on rejection by HL and ESPA1 was more substantial than NF270 and NF90, which could not be simply explained by the difference in membrane surface hydrophobicity, selective layer thickness or pore size. The HL membrane had a lower rejection of PhACs of higher hydrophobicity (log D>0) and higher HFP (>0.02). Nevertheless, the effects of PhAC hydrophobicity and HFP on rejection by ESPA1 could not be discerned. Poor rejection of certain PhACs could generally be explained by aspects of steric hindrance, electrostatic interactions and adsorption. High-pressure membranes like NF90 and NF270 have a high promise in TrOC removal from contaminated water.

Diporphyrin tweezer for multichannel spectroscopic analysis of enantiomeric excess

摘要: Chiral 1,1’-binaphthyl-linked diporphyrin ‘tweezers’ ( )- /( )- and the corresponding zinc(II) complexes ( )- /( )- were prepared as chiral host molecules, and their utility for chiral analyses (especially enantiomeric excess ( ) determinations) were evaluated. Tris(1- -dodecyl)porphyrins were used for the first time as the interacting units. Host capabilities of the diporphyrin tweezers were investigated by titrations with ( , )- and ( , )-cyclohexane-1,2-diamine (CHDA). The host molecules could be used as multichannel probes of by using UV-vis, circular dichroism (CD), fluorescence emission and H nuclear magnetic resonance ( H-NMR) methods. Chiral configurations could also be differentiated using CD or H-NMR spectroscopy. All three optical techniques give good resolution of with reasonable sensitivity considering the low concentrations used (ca. 10 mol·L ). The determination of CHDA enantiomers using NMR spectroscopy is also possible because of the reasonably well separated resonances in the case of ( , )- and ( , )-CHDA. Non-metallated ( )- /( )- hosts could not be used to detect chiral information in a strongly acidic chiral guest. This work demonstrates the utility of 1,1’-binapthyl-linked chiral hosts for chiral analysis of ditopically interacting enantiomers.
